The use of humor in my own wedding really insured that everybody kept listening properly. I used it scattered throughout my speech as a way of grabbing attention and making sure they were focusing on what I wanted to say. In any toast the bride should be the center of attention at precisely that time, so its important to get it right, and let everyone hear what you have too say.

Personally the best way to use this tactic is to inject a few jokes into your regular speech. Say emotional things, and mix it up with some humor to keep everyone relaxed and focused. I was extremely nervous and emotional on my big day, and this really helped myself as well as the audience.
